Production and Characterization of Monoclonal Antibodies to Perchloric Acid Soluble Antigen of M. tuberculosis and its Clinical Application


Abstract
Mycobacteria cause diseases which carry a considerable burden in morbidity, mortality and social problems. A battery of monoclonal antibodies specific for mycobacterial antigens would provide a useful tool for rapid diagnosis of mycobacterial
disease.
In this study, perchloric acid soluble mycobacterial antigen(TB-P) was produced, which might be thought of as a component of bacterial cell surface antigen. Antibody was tested against TB-P with the sera of patients with active pulmonary
tuberculosis
patients, by ELISA, SDS-PAGE was done with TB-P, and western blotting with the sera of patients with active pulmonary tuberculosis Monoclonal antibodies were produced against TB-P, and Western blotting was done with these monoclonal antibodies.
Indirect
Immunofluorescent staining was done against M. tuberculosis, m. bovis BCG, and pathological specimen of active pulmonary tuberculosis.
@ES Results were as follows.
@EN 1. TB-P consist of major antigens of 871, 42, 38, 10 kD and minor antigens of 54, 20, 18, 14, 12 kD proteins.
2. IgG subclass antibodies iof the sera of patients with active tuberculosis patients were IgG1, IgG2, IgG4 which reacted with low molecular antigens of TB-P, such as 14, 12, 11, 10 kI) proteins.
3. Fourteen monoclonal antibodies to TB-P were produced and were composed of IgM, IgG2a, IgG3.
4. These monclonal antibodies reacted with M. tuberculosis, M. bovis BCG, and M. tuberculosis in the pathological specimen.
5. These monoclonal antibodies reacted with 54 kD protein of TB-P.
In conclusion, TB-P and monoclonal antibodies against TB-P might be useful as a diagnostic tool of active tuberculosis.
